      目的 探讨长链非编码RNA(long non-coding RNA,lncRNA)NONHSAT005760.2在绝经后骨质疏松妇女和骨量正常妇女外周血中的表达差异,以及其在人骨髓间充质干细胞(h-BMSCs)成骨分化过程中的作用。方法 收集2019年全年在我院体检病人样本,选取30例骨密度T值≥-1 SD妇女纳入骨量正常组,30例T值≤-2.5 SD的妇女纳入骨质疏松组。病人年龄为55~70岁,平均为60岁。抽取两组病人少量外周血并提取外周血单个核细胞(PBMC)中的RNA进行lncRNA高通量测序,通过大数据分析和实时荧光定量PCR(qPCR)检测外周血PBMC细胞中lncRNA的差异表达情况。qPCR检测差异表达的lncRNA在h-BMSCs成骨分化过程中的差异表达量。结果 与骨量正常组相比,骨质疏松组病人外周血中lncRNA(NONHSAT005760.2)含量明显上调(P<0.01),同时在hBMSC诱导成骨分化过程中,NONHSAT005760.2的表达量随着诱导时间延长而降低。结论 NONHSAT005760.2在骨质疏松病人外周血中高表达,而在成骨分化过程中含量显著下降,表明其与骨质疏松密切相关,有望成为骨质疏松症的分子标志物。

      Objective To investigate the expression differences of long non-coding RNA (NONHSAT005760.2) in postmenopausal osteoporosis and normal bone mass female patients and its role in osteoblastic differentiation of human bone mesenchymal stem cells (h-BMSCs). Methods The samples of patients undergoing physical examinations in our hospital throughout 2019 were collected, and 30 women with bone mineral density T value ≥-1 SD were selected as the normal bone mass group (30 cases), and 30 women with T value ≤-2.5 SD served as the osteoporosis group. The age was 55 to 70 years old (mean 60 years old). The relative expression level of lncRNA in blood was detected by lncRNA sequencing and qPCR. qPCR was used to detect the expression of differential lncRNA during osteogenic differentiation of hBMSCs. Results As compared with normal bone mass group, the level of lncRNA (NONHSAT005760.2) in the peripheral blood of patients in the osteoporosis group was significantly increased (P<0.01), and that was increased during osteoblastic differentiation of h-BMSCs. Conclusion NONHSAT005760.2 is highly expressed in the peripheral blood of patients with osteoporosis, and its content decreases significantly during the process of osteogenic differentiation, indicating that it is closely related to osteoporosis and is expected to become a molecular marker of osteoporosis.
